Abstract

The amplification of twenty-three microsatellite loci was optimised for automatic fluorescent detection using an ALFexpress aparatus for genotyping grapevine. By optimising PCR reaction mixture and PCR condition, the generation of fragments was found to be locus specific. Problems in allele calling and sizing due to generation of additional fragments, intergel variability and allele microvariability, are also discussed.
